You probably know your blood type: A, B, AB or O. You may even know if you're Rhesus positive or negative. But how about the Langereis blood type? Or the Junior blood type? Positive or negative? Most people have never even heard of these.

Yet this knowledge could be "a matter of life and death," says University of Vermont biologist Bryan Ballif.

While blood transfusion problems due to Langereis and Junior blood types are rare worldwide, several ethnic populations are at risk, Ballif notes. "More than 50,000 Japanese are thought to be Junior negative and may encounter blood transfusion problems or mother-fetus incompatibility," he writes.

But the molecular basis of these two blood types has remained a mystery ? until now.

In the February issue of Nature Genetics, Ballif and his colleagues report on their discovery of two proteins on red blood cells responsible for these lesser-known blood types.

Ballif identified the two molecules as specialized transport proteins named ABCB6 and ABCG2.

"Only 30 proteins have previously been identified as responsible for a basic blood type," Ballif notes, "but the count now reaches 32."

The last new blood group proteins to be discovered were nearly a decade ago, Ballif says, "so it's pretty remarkable to have two identified this year."

Both of the newly identified proteins are also associated with anticancer drug resistance, so the findings may also have implications for improved treatment of breast and other cancers.

As part of the international effort, Ballif, assistant professor in UVM's biology department, used a mass spectrometer funded by the Vermont Genetics Network. With this machine, he analyzed proteins purified by his longtime collaborator, Lionel Arnaud at the French National Institute for Blood Transfusion in Paris, France.

Ballif and Arnaud, in turn, relied on antibodies to Langereis and Junior blood antigens developed by Yoshihiko Tani at the Japanese Red Cross Osaka Blood Center and Toru Miyasaki at the Japanese Red Cross Hokkaido Blood Center.

After the protein identification in Vermont, the work returned to France. There Arnaud and his team conducted cellular and genetic tests confirming that these proteins were responsible for the Langereis and Junior blood types. "He was able to test the gene sequence," Ballif says, "and, sure enough, we found mutations in this particular gene for all the people in our sample who have these problems."

Beyond the ABO blood type and the Rhesus (Rh) blood type, the International Blood Transfusion Society recognizes twenty-eight additional blood types with names like Duffy, Kidd, Diego and Lutheran. But Langereis and Junior have not been on this list. Although the antigens for the Junior and Langereis (or Lan) blood types were identified decades ago in pregnant women having difficulties carrying babies with incompatible blood types, the genetic basis of these antigens has been unknown until now.

Therefore, "very few people learn if they are Langereis or Junior positive or negative," Ballif says.

"Transfusion support of individuals with an anti-Lan antibody is highly challenging," the research team wrote in Nature Genetics, "partly because of the scarcity of compatible blood donors but mainly because of the lack of reliable reagents for blood screening." And Junior-negative blood donors are extremely rare too. That may soon change.

With the findings from this new research, health care professionals will now be able to more rapidly and confidently screen for these novel blood group proteins, Ballif wrote in a recent news article. "This will leave them better prepared to have blood ready when blood transfusions or other tissue donations are required," he notes.

"Now that we know these proteins, it will become a routine test," he says.

This science may be especially important to organ transplant patients. "As we get better and better at transplants, we do everything we can to make a good match," Ballif says. But sometimes a tissue or organ transplant, that looked like a good match, doesn't work ? and the donated tissue is rejected, which can lead to many problems or death.

"We don't always know why there is rejection," Ballif says, "but it may have to do with these proteins."

The rejection of donated tissue or blood is caused by the way the immune system distinguishes self from not-self. "If our own blood cells don't have these proteins, they're not familiar to our immune system," Ballif says, so the new blood doesn't "look like self" to the complex cellular defenses of the immune system. "They'll develop antibodies against it," Ballif says, and try to kill off the perceived invaders. In short, the body starts to attack itself.

"Then you may be out of luck," says Ballif, who notes that in addition to certain Japanese populations, European Gypsies are also at higher risk for not carrying the Langereis and Junior blood type proteins.

"There are people in the United States who have these challenges too," he says, "but it's more rare."

Ballif and his international colleagues are not done with their search. "We're following up on more unknown blood types," he says. "There are probably on the order of 10 to 15 more of these unknown blood type systems ? where we know there is a problem but we don't know what the protein is that is causing the problem."

Although these other blood systems are very rare, "if you're that one individual, and you need a transfusion," Ballif says, "there's nothing more important for you to know."

(AP) ? A western Pennsylvania woman says state environmental officials refused to do follow-up tests after their lab reported her drinking water contained chemicals that could be from nearby gas drilling.

At least 10 households in the rural Woodlands community, about 30 miles north of Pittsburgh, have complained that recent drilling impacted their water in different ways.

The Department of Environmental Protection first suggested that Janet McIntyre's well water contained low levels of only one chemical, toluene. But a review of the DEP tests by The Associated Press found four other volatile organic compounds in her water that can be associated with gas drilling.

DEP spokesman Kevin Sunday said on Friday that the low chemical concentrations were not a health risk, and suggested that the contamination may have come from the agency's laboratory itself or from abandoned vehicles on or near the property. But Sunday didn't answer why DEP failed to do follow-up tests if the DEP suspected that its own lab was contaminated.

One public health expert said the lack of follow-up tests by DEP doesn't make sense.

"DEP cannot just simply walk away," said Dr. Bernard Goldstein, professor emeritus at the University of Pittsburgh School of Public Health.

McIntyre and other residents say the water problems started about a year ago, after Rex Energy Corp. of State College, Pa., drilled two wells. But a map Rex provided also shows gas wells from other companies in the area.

Residents in the community have been complaining for nearly a year, but DEP never revealed the possible presence of chemicals to the general public.

Rex has been supplying drinking water to many households, but has sent letters notifying them it will no longer deliver drinking water after Feb. 29.

In a statement, Rex said that the wells of residents who have complained are from 2,100 to 4,600 feet from its drilling locations. The company noted that many other homeowners in the area haven't raised complaints or concerns.

Rex also said there are old oil wells in the region that could impact some ground water, and that there were "no notable differences in water chemistry between pre- and post-drill water quality tests of the water wells in question."

McIntyre's water showed detectable levels of t-Butyl alcohol, acetone, chloromethane, toluene and 1, 3, 5-trimethylbenzene. The chemicals can be used in the high-pressure hydraulic fracturing process that has led to a production boom of deep shale gas in Pennsylvania. But some are also commonly used in households and other industry, such as toluene, a paint thinner.

Goldstein said the multi-chemical mix is what is so unusual, since it suggests either multiple sources of contamination, or an industry that uses many different chemicals.

"Where would you get such a strange mixture?" Goldstein asked. "Is this coming from drilling?" He added that the low concentrations shown in the test may not be a health threat, and may not be connected to gas drilling. But if DEP's own laboratory was even a potential source of the chemicals, the agency had the obligation to follow up.

"You've got to pursue the finding," Goldstein said, since if the lab was at fault the variety of chemicals that showed up "makes no sense at all, except a really sloppy lab."

Sunday said an independent peer review of the DEP laboratory found it to be "a well-managed, efficient and highly functional laboratory" that is "driven by a culture of customer service."

McIntyre told the AP that she repeatedly asked a DEP field worker for follow-ups after two separate tests last summer showed the chemicals, as well as elevated levels of some natural underground compounds such as barium.

"He said no," she said, leaving her feeling that she had no one to turn to for an objective public health opinion. She also said the chemicals didn't show up on pre-drill water tests.

As drillers have poured into Pennsylvania to tap its vast Marcellus Shale gas reserves, residents and environmentalists have raised concerns over the impact or potential impact to water supplies. Water contamination in Dimock, in northeast Pennsylvania, has riled some homeowners for months.

State regulators determined that Houston, Texas-based Cabot Oil & Gas Co. drilled faulty gas wells that allowed methane to escape into Dimock's aquifer. The company paid heavy fines but denied responsibility; it has been banned from drilling in a 9-square-mile area of Dimock since April 2010.

Another Woodlands resident who complained about dramatic changes in her water over the last year said DEP staff suggested the bad smell was simply from garden slugs in her well, which is 300 feet deep.

"They just insult your intelligence. I don't trust the DEP," said Kim McEvoy, who lives about a mile from McIntyre.

McEvoy said she wants the U.S. Environmental Protection Agency to investigate the community, and that she's come to that point because state environmental officials haven't answered her questions.

"Something has happened here," McEvoy said.

How to Read a Patent Like a Pro - IP Law For Startups

There is a fine art to reading patents. And it?s far harder than you might imagine.

The devil is really in the details.

Today, I?m explaining how to read a patent like a pro, including identifying the most important parts and considerations for interpretation. My primary goal is to help you get a better understanding of how to determine what a patent actually covers.

Frequently, it?s not what you?d initially think.

Part 1: ?Patent Anatomy

Patents have several common sections:

  • the front page basics including the patent number, title, inventor?s name, owner if different, patent filing date, patent issue date, and cited references.
  • the abstract, which gives a snapshot of the type of invention and what it does;
  • the specification, which explains how to make and use the invention;
  • the drawings, which illustrate the invention; and
  • the claims, which define the legal boundaries of the patent?s coverage.

For example, see Patent No.4,833,729, a patent for a Shark Protector Suit.

From the front page, you can get a general idea about what the invention does. Here, the abstract describes the invention as a rubber suit with spikes ?to prevent a shark from clamping its jaws over a wearer.?

But if you want to read a patent like a pro, after quickly reading the title and the abstract, you need to start at the end of the patent ? with the claims.

The claims are the most critical part of a patent.

It is the claims as issued that determine the scope of the patent?s reach. And the claims may have undergone major modifications during the patent prosecution process with the PTO. The specification may describe a general invention that seems to have broad coverage. Indeed, the initial claims may have been very broad and far reaching ? too far reaching. Because of conflict with prior art, the inventor may have significantly narrowed what is being claimed to get the patent granted. And the issued patent may not even cover some of the ?preferred embodiments,? examples of the invention described in the specification.

Don?t be fooled by broad language in the specification. You need to first look at the claims to get a sense of what is actually covered.

Then, you need to try to figure out what the claim language actually means ? the claim construction.

Part 2: ?Claim Construction for Beginners

Claims come in two flavors: ?1) independent, stand alone claims; and 2) dependent, me-too type claims that incorporate an earlier claim with an additional element.

Claim construction is all about the meaning of individual words and phrases in the claims. Don?t be thrown off by the awkward wording or use of the term ?said.? Drafters are trying to define the invention clearly and doing so can make the wording strange.

A definitive claim construction can be hard, very hard. Indeed, according to the Federal Circuit Court of Appeals, even district court judges in patent infringement suits get the claim construction wrong around half of the time. (In patent cases, the judge decides the claim construction after briefing by the attorneys, who try to persuade the judge of their particular interpretation. See example of a judicial claim construction analysis.) Moreover, millions of dollars are paid to attorneys who fight over the meaning of a few claim words. Amazingly nuanced, claim construction can be a high stakes squabble with billions of dollars in damages hinging on the meaning.

So how do you determine what a patent?s claim terms mean, especially when you?re not a lawyer?

First, start with the plain meaning of the words. How are they generally understood by a person skilled in the art? How are they defined in a standard or technical dictionary? Is there any ambiguity as to the meaning?

Second, read the specification very, very carefully. Have certain terms been coined or defined by the inventor? Examine the drawings and examples for clues about the meaning of the terms.

Third, and most importantly for infringement considerations, review the prosecution file history ? the paperwork between the PTO and inventor as the patent application was examined. (You can find it on-line). The inventor may have had to distinguish the invention from prior art. She may have had to tell the examiner what the invention is not. ?These limitations can be critical. The inventor cannot go back and try to insist that the invention covers what was disclaimed, although I?ve seen some plaintiff?s who have tried.

From these sources, you will have a basic idea about what the claims mean and what is covered or not covered.

My SO and I have quite different views on an individual's right to privacy within a relationship, and I know when I've read similar topics on here there have been diverging opinions.

Tonight he accused me of "withholding intimacy" because I asserted (what I believe to be) my right to my privacy. A few nights ago he had asked me what I was reading on my e-reader and I said it was a self-help book, without going into further detail -- it was a book about emotional unavailability. I avoided telling him specifically which book it was because in the past he has immediately personalized the subject ("Oh, so you think I'm emotionally unavailable??") and has also mocked me quite nastily for reading self-help books. I did explain this to him at the time and he denied ever doing it -- he says I must have misunderstood him. There have also been times that he has criticized the novels that he's seen me read, saying that they're "stupid". So I'm a bit wary of revealing my reading material!

Bottom line, unless I'm reading a manifesto on how to murder one's SO and escape undetected , I don't see why it matters what I'm reading. Normally he can just glance at the cover of the book and know the answer in a second; he doesn't have that advantage with the e-reader.

So tonight he started in on me about being secretive and said he had just been making conversation, whereas I was "rude and hostile" by refusing to anwer his question about the book I was reading. I explained that, in a general sense, people have the right to not provide information according to their own boundaries -- he seems to feel that posing the question obligates the other person to answer, that he is somehow entitled to have his curiosity satisfied. The same if he asks me "What are you thinking about?".

He got pissed off and walked out. Over this.

My question: how much privacy can an individual reasonably expect within a relationship?

Secondly, this is not a question of privacy or boundaries. You weren't on the toilet. If your husband asks what you are doing on the internet or what you're reading, you should be willing and forthcoming with the answers. Otherwise, you could quite easily begin issues that are matters of trust. The last thing you want is to end up wondering and feeling insecure about who he is talking to or whose blogs he's reading. That's where your "privacy and boundaries" is headed. And when it all starts, you won't possess the presence of mind to remember you are the one who started that mess. Moreover, you called yourself protecting yourself and preventing an argument, but an argument ensued anyway, didn't it? And you weren't so protected afterall.

First and foremost, he makes conversation by asking what you are reading, not by criticizing what you read and thereby (and quite insultingly), criticize you and the reading material. However, what he does (and says) is not nearly so important as what you do about what he does (and says). You took it from him and allowed him to judge, belittle, and berate you, and then you allowed him to entirely dismiss your expressions and feelings about his insults by placing the blame, the offense, and the breakdown in communication squarely on your shoulders. He stormed out of the room like a wounded child. In other words....according to him....you offended yourself AND him. What kind of mess is that?

Your solutions were to first hide the subject matter of your reading material so as to prevent him belittling you over it, and then you submitted to an entirely different (and pointless) argument about privacy and boundaries. You will forever flip, flop, and fly around in the dust of his wrath if you keep getting caught up like this and not stand up for yourself.

The polite and engaging thing for him to do (since he so sincerely CLAIMS he was only making conversation) would have been to further inquire of your reading material in such a way that he proved himself considerate, interested, and engaging (in conversation). You know, he could have shown a little interest and possibly improve himself by it. Honestly though, I don't know what you are learning if this is example of how you handle things with your husband. Stand up for yourself and rather than constantly trying to sneak and hide to protect yourself from his judgmental and offensive nature, stop him in his tracks when he offends you.

if you are not married or living together and he is being this way, ask yourself, is this the kind of relationship you you really want? it doesnt sound like it, which is fine, but you should move on that.

I've tried that in the past and he uses the old "I was only joking" defense or he repeats his exact words in a pleasant, neutral tone instead of the actual mocking tone in which it was first delivered and asks "What's wrong with saying that?" (well, duh, tone is everything!). Then tells me I'm being overly sensitive.

I'd have to say that the biggest problems we've been having in the relationship lately are due to the fact that I am stopping him in his tracks more often instead of being the compliant little doormat that he was used to.

Job Growth in Gaming Industry Slowed by Massive Debt

LAS VEGAS -- In the mid-1980s, before the era of megaresorts, Nevada's gaming industry generated roughly $3 in revenue for every dollar spent on long-term debt.

As the public began craving bigger and newer resort attractions, and gaming companies did whatever they could to meet demand, the construction debt began piling up. Add the most expensive company buyout in gaming history, coupled with the ravages of the Great Recession, and the result today is a gaming balance sheet with more debt than revenue.

The authors of two separate studies on casino finances say massive debt slows the economy because money that could be spent hiring more workers goes instead to pay off loans. As David Schwartz, director of the UNLV Center for Gaming Research, said Thursday, that translates to poorer customer service if it means a hotel could have had five workers at their registration desk instead of four.

"If you've got more money going toward debt and interest payments, that's less money you can spend on other things," he said.

Neither Schwartz or Jeremy Aguero, a principal at Las Vegas economic analysis firm Applied Analysis, say they believe gaming companies have dug a hole so deep they cannot recover. To the contrary, there are signs the industry's recovery is under way.

"We will go over 40 million visitors this year, which will be a record, and I think it will be a real big deal," Aguero said.

Late last month, Schwartz reported that Nevada's long-term gaming debt last fiscal year was nearly $24.5 billion plus $3 billion in interest, versus $22 billion in revenue. Aguero reported in 2010 that Nevada gaming debt exceeded revenue for the first time in fiscal 2008. That's when the industry owed $1.22 for every dollar taken in.

A major contributor to the debt that year was the $17.7 billion buyout of Harrah's Entertainment -- now Caesars Entertainment -- by private equity firms Apollo Management and Texas Pacific Group. Companies also ran up massive construction debt led by the likes of MGM Resorts International, a partner in the $8.5 billion CityCenter project that remains $2.5 billion in debt.

Gaming revenue is once again on the rise, a trend that will continue if Aguero's prediction of record visitation to Las Vegas holds up. As long as revenue continues to increase, Aguero said he believes lenders will extend the maturation dates on gaming loans, which many already have done. Aguero said the industry already has begun paying down principal, with last year's $24.5 billion debt representing a $3.9 billion drop from the peak in 2008.

Station Casinos ended last year with $2.5 billion in debt, a considerable drop from a $5.9 billion debt when the company filed for bankruptcy in 2009.

Credit rating agencies are also beginning to look more favorably at the industry, which can lead to better lending conditions for the companies. Moody's Investors Service announced earlier this month that it would review Caesars Entertainment's debt for a possible upgrade. Fitch Ratings last fall upgraded MGM Resorts debt and Moody's did the same last year for Las Vegas Sands Corp.

Gaming, though, is not completely home-free. The Cosmopolitan announced Tuesday that it is laying off more than 40 workers. And the days of constructing new multi-billion-dollar megaresorts on the Strip are still over for the foreseeable future.

Gaming companies with substantial debt can survive if they become smarter with their money, Schwartz said.

"Casinos have to be a bit more cautious because of their debt," he said.

That means settling for smaller-scale projects and entering more business partnerships to avoid adding to the debt.

He cited an example of both: Caesars Entertainment is pressing forward with The Linq, a $550 million retail development that will include a 550-foot observation wheel; and MGM Resorts announced Wednesday it would partner with Ameristar Casinos to extend offers to each other's players.

"Ten years ago, MGM would have tried to buy Ameristar," Schwartz said.
